What is Zopiclone?

Zopiclone belongs to the group of medicines called Central Nervous System (CNS) depressants. Because of its calming effect on the brain, Zopiclone is commonly used to treat insomnia. It helps you fall asleep more quickly, improves the quality of your sleep and stops waking up during the night or too early in the morning. Zopiclone also helps in case of severe problems of sleep due to mood or mental health problems.

Zopiclone is ‘’non benzodiazepine’’ agonist of benzodiazepine receptors. It also has a sedative, anxiolytic, muscle-relaxing center, anticonvulsant and amnesic properties. Zopiclone enhances GABA-ergic processes in the brain, interacting with the benzodiazepine receptors, as a result of the increased sensitivity of GABA-receptors to the neurotransmitter.

Being a derivative of benzodiazepine, Zopiclone shares an almost identical pharmacological profile to them. Zopiclone interacts with other parts of the benzodiazepine receptor than benzodiazepines. It acts as a full agonist, which in turn positively modulates benzodiazepine-sensitive GABAa receptors and enhances GABA binding at the GABAa receptors to produce Zopiclone’s subjective effects. Since GABA functions as the brain’s predominant inhibitory neurotransmitter, such activation of receptors results in the sedative and anxiolytic effects of Zopiclone.

Zopiclone generic and brand name

Zopiclone is also known under the brand names, such as: Zimovane, Imovane, Ran-Zopiclone, Rhovane Zimovane, Zileze, Zimoclone, Zopitan, Zorclone.

Why Zopiclone prescribed?

Zopiclone is recommended for the short-term treatment of insomnia. Your body can get used to it very quickly, and after some time it is unlikely to have the same effect. Your body can also become dependent on it. That is why Zopiclone is recommended for the short-term treatment of transient, situational and chronic insomnia characterized by difficulty in falling asleep, frequent nocturnal awakenings and early morning awakenings.

How Zopiclone works

Zopiclone belongs to the class of medications called sedative-hypnotics. Do not take Zopiclone when a full night’s sleep is not possible or before you would need to be active and functional. Wait at least 12 hours after taking this medication before driving or engaging in other activities that require mental alertness. Your body needs time to eliminate the medication from your system, otherwise, impaired judgment and memory lapses may occur. If you have been taking this medication regularly for an extended period of time, do not stop taking it suddenly without talking with your doctor.

Recommended doses of Zopiclone

Many things can affect the dose, such as body weight, other medical conditions, and other medications. The usual starting dose is 3.75 mg orally at night and it takes around 1 hour to work. That dose is recommended for elderly patients or if a patient has kidney or liver problems. Taking a lower dose in these cases reduces the risk of excessive sleepiness and other side effects. The recommended adult dose of Zopiclone is 7.5 mg. The maximum daily dose is 5 mg for seniors, people with reduced liver or kidney function, and people taking certain medications. In cases of severe insomnia the dose can be increased up to 15 mg. Please, bear in mind that this medication may be habit-forming and should be taken exactly as prescribed by your doctor.

What are the pharmacokinetic properties of the drug?

Zopiclone is rapidly absorbed, with a bioavailability of approximately 80%. The peak plasma concentration of 60 ng/mL is achieved within 90 minutes. The plasma protein binding of zopiclone has been reported to be between 45 and 80%. Zopiclone is partly metabolized in the liver to form an inactive N-demethylated derivative and an active N-oxide metabolite. Approximately 50% of the administered dose is decarboxylated and excreted via the lungs. Less than 7% of the drug is also excreted in unchanged form.
